Controlled Light Exposure Microscopy (CLEM): Photobleaching and Phototoxicity Reduced

چکیده

Controlled Light Exposure Microscopy (CLEM): Photobleaching and Phototoxicity Reduced Photobleaching and phototoxicity are often the Achilles‘ heels of fluorescence live-cell imaging. Controlled Light Exposure Microscopy (CLEM) is a novel, simple imaging approach that reduces photobleaching and phototoxicity 2 to 10-fold without compromising image quality. The basic concept of CLEM is that light is only used where it is needed by spatial control (pixel by pixel) of the light exposure time. Experiments show that CLEM results in a 2 to 10-fold reduction of photobleaching and phototoxicity. As a bonus, CLEM increases the dynamic range of the fluorescence intensity at least by a factor of two.

Stroboscopic illumination using light-emitting diodes reduces phototoxicity in fluorescence cell imaging.

Excited fluorophores produce reactive oxygen species that are toxic toward many live cells (phototoxicity) and accelerate bleaching of the fluorophores during the course of extended or repeated measurements (photobleaching). We recently developed an illumination system for fluorescence microscopy using a high power light-emitting diode (LED), which can emit short pulses of light (0.5-2 ms) to e...

Programmable Array Fluorescence Lifetime Spectroscopy/microscopy Using Tcspc

Spatial light modulators (SLMs) are a family of optical devices that allow modulation of intensity and/or phase in light microscopy to control excitation and emission. There are broadly two classes of devices employed: digital micromirror arrays (DMDs) and ferroelectric liquid crystal devices. DMDs have found use in controlled light exposure microscopy (CLEM) to reduce photobleaching and photot...

Scanned LSFM with Confocal Detection

Light sheet fluorescence microscopy (LSFM) is a technique with a vivid development [1, 2]. In contrast to epi-illumination microscopy the sample is illuminated with a thin light sheet orthogonally to the detection path. Most often the sheet is formed by rapidly scanning a laser beam across the sample. This illumination yields intrinsic optical sectioning and reduction of photobleaching and phot...

Label-free intracellular transport measured by spatial light interference microscopy.

We show that applying the Laplace operator to a speckle-free quantitative phase image reveals an unprecedented level of detail in cell structure, without the gradient artifacts associated with differential interference contrast microscopy, or photobleaching and phototoxicity limitations common in fluorescence microscopy. This method, referred to as Laplace phase microscopy, is an efficient tool...
